PREPARATION

A.

Prepare
37-38 degree Centigrade water baths. Two large styrofoam cups with four
inches of water work well.
|
B.
|
Warm
insemination rod, syringe, and needle to 37-38 degrees Centigrade in an
incubator. DO NOT SUBMERGE IN WATER
BATH!
|
THAW
PROCEDURES
|
|
1.
|
Open
shipping tank. Locate aluminum cane labeled "Thaw media". Remove
small vial containing thaw media and warm to room temperature. Estimated
time: 10-15 minutes.
|
2.
|
Put
1/2 cc. of room temperature THAW MEDIA in each culture tube. (One culture
tube with 1/2 cc thaw media for each straw.) Place tubes in 37-38 degree
centigrade water bath. Keep these tubes at 37-38 degrees centigrade THIS
IS IMPORTANT!

3.
|
Have
37-38 degree centigrade water bath ready for thawing semen.
WARNING! REMOVE STRAW ONLY WHEN YOU ARE READY TO THAW! Each semen straw must be thawed at 37-38 degrees C. Monitor temperature carefully for each straw. |
4.
|
Use
tweezers or hemostats and hold straw at tip of open end. Submerge in 37-38
degree C. water bath. Do not let water get into the open end of the straw.
|
5.
|
Remove
straw when air bubbles move freely when you invert straw. This usually
takes 10-15 seconds.
|
6.
|
Dry
straw completely on paper towel immediately.
|
7.
|
Do
subsequent straws using same method.
|
8.
|
Hold
straw open end over the warmed 37-38 degree C. Thaw Media in the glass
culture tube. Cut straw just below cotton plug. Semen will run into the
Thaw Media.
|
9.
|
Let
combined thawed semen and thaw media stand in the glass culture tube for
2 minutes in the 37-38 degree C water bath.
|
10.
|
Draw up with large bore needle or insemination rod. Immediately inseminate into bitch. |
